摘要 目的:建立一测多评法同时测定陈皮中6个黄酮类成分的含量。方法:采用OSAKA CAPCELL PAK C18色谱柱(250 mm×4.6mm,5μm),流动相为乙腈-0.1%甲酸溶液,梯度洗脱;流速为1.0 mL?min-1,柱温为30°C,检测波长为330 nm。以橙皮苷为内参物,计算橙皮苷与芸香柚皮苷、香蜂草苷、川陈皮素、3,5,6,7,8,3′,4′-七甲氧基黄酮、橘皮素之间的相对校正因子,分别采用一测多评法和外标法对15批陈皮中6个黄酮类成分的含量进行测定并比较结果。结果:6种成分在各自范围内线性关系良好(r≥0.9999);平均加样回收率99.95%~102.97%,RSD 0.81%~2.03%。一测多评法与外标法的含量测定结果无显著性差异(P>0.05)。结论:该方法操作便捷、准确可靠、重复性好,可用于陈皮的质量控制。 |

Abstract objective: To establish a method for determination of six Flavonoids in Citri Reticulatae Pericarpium by quantitative analysis of multi-components by single marker. Methods:A OSAKA CAPCELL PAK C18 column was used(250 mm×4.6mm,5μm), with acetonitrile -0.1% formic acid as mobile phase with gradient elution, The flow rate was set at 1.0 mL·min-1,the temperature maintained at 30 ℃,and the detection wavelength was set at 330 nm. Hesperidin was chosen as the internal reference substance to caculate the relative correction factors(RCFs)of narirutin,didymin,nobiletin,3,5,6,7,8,3"",4""-heptamethoxyflavone,tangeretin. Using relative retention time method and external standard method(ESMS)to determinate the contents of 6 Flavonoids in 15 batches of Citri Reticulatae Pericarpium and compare the results. Results:The linear relationships of six component were well (r>0.9999). The average recoveries were 99.95%~102.97% with RSDs of 0.81%~2.03%. There were no significant differences(P>0.05) between the calcurlated value and estimated value on QAMS and external standard method of content determination. Conclusions:The method is accurate and reliable,sensitive,reproducible and easy to operate. and can be applied to the determination of Citri Reticulatae Pericarpium and control the quality. |
