目的 建立双波长HPLC法测定小儿感冒颗粒中绿原酸、木犀草苷、3,5-O-二咖啡酰基奎宁酸和连翘苷的含量。方法:Agilent C18 色谱柱(4.6mm╳250mm,5μm);流动相:乙腈-0.4%磷酸,梯度洗脱;检测波长:0-45min(327nm),45-83min(202nm);柱温:30℃;流速:1mL/min,进样量10μL。结果:绿原酸、木犀草苷、3,5-O-二咖啡酰基奎宁酸、连翘苷浓度分别在 0.01267~0.3801μg/m L、0.1040~1.5600μg/mL、0.00840~0.2526μg/mL、37.90~319.021μg/mL范围内呈良好的线性关系(r=0.9999,1.0000,0.9966,1.0000);平均加样回收率分别为97.89%、96.37% 、96.63%和103.3%(n=6)。结论:所建立的双波长HPLC测定法简便、准确、重复性好、专属性强,可同时测定小儿感冒颗粒中的多种有效成分的含量。 |

Abstract Objective: To establish a method for determination Chlorogenic Acid、Luteolin、3,5-O-Dicaffeoyl Quinic Acid and Forsythin in Xiaoerganmao Granules by dual-wavelength HPLC.Methods: Agilent C18 (4.6 mm x 200 mm, 5 μm) .The mobile phase was acetonitrile-0.4% phosphate with gradient elutionthe. The column temperature was 30℃.The detection wavelength was set as 327 nm (Chlorogenic Acid、Luteolin、 3,5-O-Dicaffeoyl Quinic Acid),202 nm (forsythin) and the flow rate was 1.0 mL ? min-1 .Results:The linear ranges of Chlorogenic Acid、Luteolin、3,5-O-Dicaffeoyl Quinic Acid and Forsyhin were between 0.01267-0.3801μg?m L-1 (r=0.9999), 0.1040-1.5600μg?m L-1 mL(r=1.0000),0.0084
0-0.2526μg?mL-1 (r=0.9966),0.03190-0.3190μg?m L-1 mL(r=1.0000).The average recoveries (n
= 6) were 97.89%,96.37%, 96.63%,103.3%,respectively.Conclusion:The method is simple,accurate and reproducible,and it can be used for quality control of Xiaoerganmao Granules . |
