      目的:建立茅莓中表儿茶素的高效液相色谱含量测定方法。方法:样品以50%甲醇溶液超声30分钟,6000 rpm离心3分钟,取上清液过0.45μm的微孔滤膜后进行HPLC分析。采用Alltima C18(250 mm×4.6 mm, 5 μm)色谱柱,柱温为35℃;流动相为0.01 mol.L-1枸椽酸-甲醇(80:20),流速为1.0 mL.min-1;检测波长为280 nm。结果:表儿茶素浓度在0.08~2.22 μg.mL-1范围内呈良好的线性关系(r=0.9999);平均回收率在99.12%;RSD=1.29%。结论:本方法灵敏、准确,具有良好的重复性和稳定性,可用于茅莓中表儿茶素的含量检测。
英文摘要:
      Objective:To develop an HPLC method for determination of epicatechin in Rubus Parvifolius L.F. Methods:The samples were extracted with 50% methanol solution for 30 minutes and centrifuged at 6000 rpm for about 3 minutes, and then the supernatant was filtered used the micropore membrane of 0.45μm for HPLC. HPLC analysis was performed on a Alltima C18 (250 mm× 4.6 mm ,5 μm). The mobile phase was 0.01 mol.L-1 citric acid - methanol (80:20) at the flow rate of 1.0 mL.min-1. The detection wavelength was 210 nm and the column temperature was 35 ℃. Results:epicatechin presented good linearity in the ranges of 0.08~2.22 μg.mL-1 (r=0.9999). The average recovery was 99.12% with RSD of 1.29%. Conclusion:The method is sensitive and accurate with good repeatability and stability, which is useful for the determination of epicatechin in Rubus Parvifolius L.F.
