      目的: 建立猴枣药材中鞣花酸含量测定方法,并对猴枣不同部位中鞣花酸的含量进行测定。方法:采用Agilent ZORBAX SB-C18色谱柱(4.6 mm×150 mm,5 μm);流动相为0.1%甲酸水-乙腈(83:17)等度洗脱;流速1.0 mL?min-1;检测波长254 nm;柱温:室温。结果: 鞣花酸的质量浓度与峰面积在测定范围内均呈良好的线性关系,其标准曲线及线性范围为 Y =76.566X+89.62(0.0495 ~989.4 μg?mL-1,r=0.9998);平均加样回收率为99.4%,RSD为1.2%。3批猴枣及其不同部位样品鞣花酸含量结果:平均含量为65.0%,其外层的鞣花酸平均含量最高,为91.0%,而中层、种仁鞣花酸平均含量较少,分别为4.3%、0.6%。结论 该方法简单,准确,重复性好,具有良好的稳定性,可用于猴枣药材质量分析和控制。
英文摘要:
      Objective: To establish a method for the simultaneous determination ofEllagic acid in Phyto Bezoar by HPLC,andestablish the method for the content determination in differentparts of Ellagic acid in Phyto Bezoar. Methods: HPLC was performed on Agilent ZORBAX SB-C18 column (4.6 mm×150 mm,5 μm) with the column temperature of 28℃; The mobile phase was 0.1% formic acid-acetonitrile (83:17) in a isocratic elution at a flow rate of 1.0 mL?min-1;The detection wavelength was set at 254 nm. Results: There were good linear relationships between the logarithm values of concentrations and those of the peak areas of the Ellagic acid, The calibration curves and linear range of the Ellagic acid was determined as Y =76.566X+89.62 (0.0495 ~9894 μg?mL-1, r=0.9998); The recoveries of the Ellagic acid was 99.4%, the relative standard deviations (RSDs) was 1.2%. The contents of the Ellagic acid showed stable among the 3 batch number tested samples, but showed different in 3 parts of Phyto Bezoar ranged from 0.6% to 91.0%. Conclusion: The established method is rapid, accurate with high repeatability, which could be used as the content determination method of the Ellagic acid in Phyto Bezoar from differentparts.
