      测定和比较桑白皮药材和黄酮提取物中桑根酮C、D和桑辛素的含量。方法:薄层色谱法鉴别药材,用硅胶G为薄层板,三氯甲烷-甲酸-甲醇( 5∶ 0.3∶1) 为展开剂,置于365nm紫外灯下检视斑点;紫外分光光度法检测桑白皮药材和黄酮提取物中总黄酮的含量;高效液相色谱法测定桑白皮药材和黄酮提取物中桑根酮C、D和桑辛素的含量。结果:薄层色谱鉴别中,桑根酮D的荧光斑点清晰;紫外分光光度法中桑根酮C在0.0391~0.1564mg/mL与吸光度呈良好线性关系( n = 5) ,平均回收率为96.7%,RSD 为1.49% ( n = 5) ;高效液相色谱法中,桑根酮C、D 及桑辛素在进样量范围内线性关系良好,桑根酮C、D 、桑辛素线性范围分别为0.185~1.48μg、0.086~0.43μg、0.233~2.097μg,平均回收率分别为97.09%、 98.2 %、 97.3%,RSD分别为1.85 %、2.32 %、1.55%(n=6)。结论:该方法简单有效,可准确测定桑白皮药材和黄酮提取物中有效成分的含量。
英文摘要:
      Objective: To determine and compare the contents of sanggenons C,D and morusin in Mori Cortex and Cortex Mori flavone extracts.Method: TLC method was used to identify sanggenon D in the sample with silica gel G plate and a mixture of chloroform-methanol-formic acid( 5∶ 1∶ 0. 3) as a developing solvent. UV was used for determination the centent of total flavonoids ofMori Cortex and Cortex Mori flavone extracts with sanggenon C as the reference substance. HPLC was applied for determination the contents of sanggenons C,D and DNJ of Mori Cortex and Cortex Mori flavone extracts.Results: In the TLC chromatogram,sanggenon D showed a distinct fluorescence spot under UV 365 nm with good separation. In UV,sanggenon C calibration curve showed a good linear relationship at the range of 0.0391~0.1564mg/mL, the average recovery was 96.7% ,RSD = 1.49%(n=5) ; For the HPLC quantitation method,sanggenons C,D and morusin showed good linear within the scope of 0.185~1.48μg、0.086~0.43μg、0.233~2.097μg,respectively. The average recovery of sanggenons C,D and morusin were 97.09%(RSD=1.85 %)、 98.2 %(RSD=2.32 %)、 97.3% (RSD=1.55%)(N=6),respectively.Conclusion: The method is simple and effective, and can accurately determine the contents of sanggenons C,D and morusin in Mori Cortex and Cortex Mori flavone extracts.
