      目的:优化枸杞子中甜菜碱的HPLC-UV含量测定方法。方法:采用甲醇提取,筛查碱性氧化铝与洗脱溶剂用量优化前处理的制备。采用XBridge Amide色谱柱(4.6×250mm,3.5um),以乙腈-水(85:15)等度洗脱;流速为1.0 mL/min;检测波长为195 nm;柱温30 ℃。结果:前处理制备方法为用甲醇加热回流提取1小时,过7g碱性氧化铝柱,用乙醇100ml洗脱。甜菜碱进样量在0.2376~5.9406 ug(r=0.9999)范围内线性关系良好,平均加样回收率(n=9)为97.42%(RSD=1.39%)。结论:该方法简便、可靠,重现性好,目标峰干扰少,可以用于枸杞子中甜菜碱的定量分析,并与药典方法对枸杞子中甜菜碱的含量测定结果进行了比较,可以用于现行标准的改进。
英文摘要:
      Objective To optimize the method for the determination of betaine in Lycii Fructus by HPLC-UV. Methods Methanol was used to extract the betaine. The effects of alkaline alumina quantity and the elution solvent volume were examined to optimize the preparation method. The analyses was performed on a XBridge Amide column(250 mm×4.6 mm, 3.5 μm), mobile phase was acetonitrile- water(85:15) with isocratic elution mode at the flow rate of 1.0 mL.min<sub>-1</sub>; The detection was set at 195 nm, the column temperature was 30 ℃. Results Optimal preparation method was heat under reflux for 1 hour, apply to a column packed with 7g alkaline alumina, elute with 100ml ethanol. The results showed good linearity in the ranges of 0.2376~5.9406μg(r=0.9999). The average recovery (n=9) was 97.42% with the RSD was1.39%. Conclusion This method is simple, reliable and reproducible, the target peak is less loss, which is suitable for the determination of betaine in Lycii Fructus. The results of this method was compared with the Pharmacopoeia method. This method can be used to provide a basis for the quality control of Lycii Fructus.
