目的:建立测定复方芩兰口服液中绿原酸、咖啡酸、黄芩苷、连翘苷和汉黄芩苷的含量HPLC分析方法。方法:采用色谱柱Agilent Eclipse plus C18(4.6×150mm,3.5μm),流动相为甲醇-10%冰醋酸梯度洗脱;绿原酸和咖啡酸检测波长为324nm,黄芩苷、连翘苷和汉黄芩苷检测波长为274nm;流速为1.0mL/min;柱温为40℃,采用液质联用法鉴定五个组分。结果:绿原酸、咖啡酸、黄芩苷、连翘苷和汉黄芩苷五种成分在测定的范围内均呈现出良好的线性关系(r≥0.9998),平均回收率为97.80%~101.94%(n=6)。结论:该方法操作快速简便,分离效果好,灵敏度高,重现性良好,为测定复方芩兰口服液主成分的含量提供了一种快速高效可靠的方法。 |

Objective:To establish a HPLC method for the determination of chlorogenic acid, caffeic acid, baicalin, forsythin and wogonoside in Compound QinlanOral Solution. Methods: The Agilent Eclipse PlusC18column(4.6 × 150 mm, 3.5 μm) was used .The mobile phase was gradient elution with methanol-10% acetic acid at a flow rate of 1.0mL / min. The detection wavelength of chlorogenic acid and caffeic acid was 324 nm and the detection wavelength of baicalin Forsythin and wogonoside was 274nm. The column temperature was 40 ℃. Five components were identified by LC/MS/MS.Results: The five compounds (chlorogenic acid, caffeic acid, baicalin, forsythin and wogonoside) showed a good linear relationship (r≥0.9998) and the average recoveries was 97.80% -101.94%(n=6). Conclusion: The method is simple, rapid, sensitive and reproducible. It provides a fast, efficient and reliable method for the determination of the main components of Compound QinlanOral Solution. |
