摘要 目的:建立浊度法测定制霉素片含量的方法。方法:以白色念珠菌为试验菌,加菌量0.3%(每100mL培养基中加入0.3mL试验菌),35℃培养3~4h,530nm波长处在线测定其吸光度。结果:在4.0~12.0U?mL-1的范围内, 吸光度(A)与浓度对数(lgC)呈良好线性关系,平均回收率为98.9%(RSD=1.2%),浊度法与现行标准管碟法测得的含量结果基本一致。结论:浊度法可代替管碟法作为制霉素片含量的测定方法。 |

Objective: To develop a turbidimetric method for the content determination of Nysfungin Tablets. Method: Candida albicans was the test organismsand adjusted to 0.3% ratio (0.3mL test bacteria were added to each 100mL medium) in spore suspension.Absorbance was determined on-line at 530nm after an incubation period of 3-4h at 35℃. Results: In the range of 4.0 ~ 12.0U?mL-1, the absorbance (A) and the concentration logarithm (lgC) showed a good linear relationship, the average recovery was 98.9% with RSD of 1.2%. The result of turbidimetric method was consistent with the result of current standard cup-plate method. Conclusion: Turbidimetric method can replace cup-plate method to determine the content of Nysfungin Tablets. |
